Purified centrosomes are assayed in vitro for microtubule nucleation activity. The centrosomes are stained using γ-tubulin specific antibody (green); the microtubules are stained using an antibody specific for α-tubulin (red). The centrosomes appear yellow since they are positive for both markers. The microtubule content of the aster (star shaped structure of microtubules emanating from the centrosomes) directly correlates with microtubule nucleation function of centrosomes. The BRCA1 E3 ubiquitin ligase inhibits centrosome function by targeting proteins that attach γ-tubulin to the centrosome. For a full discussion, see the communication by S. Sankaran, D.E. Crone, R.E.
